  Abstract Due to its role in aging and antitumor defense, cellular senescence has recently attracted increasing interest. However, there is currently no single specific marker that can unequivocally detect senescent cells. Here, we identified I2-L-fucosidase (I2-Fuc) as a novel sensitive biomarker for cellular senescence. Regardless of the stress stimulus and cell type, I-Fuc activity was induced in all canonical types of cellular senescence, including replicative, DNA damage- and oncogene-induced senescence. Strikingly, in most models the degree of I-Fuc upregulation was higher than the induction of senescence-associated I2-galactosidase (SA-I2-Gal), the current gold standard for senescence detection. As I-Fuc is convenient and easy to measure, we suggest its utility as a valuable marker, in particular in cells with low SA-I2-Gal activity.

  Title Behavioral and plasma monoamine responses to high-speed railway noise stress in mice Type Journal Article

  Abstract Studies have reported that railway noise causes stress responses. To evaluate the effects of high-speed railway (HSR) noise on behaviors and plasma monoamines. Institute of cancer research mice were exposed to previously recorded HSR noise for 53 days. The noise was arranged according to the HSRs 24-h traffic number and adjusted to a day-night equivalent continuous A-weighted sound pressure level (Ldn ) of 70 dB (A). The open field test (OFT) and the light/dark box test were applied to observe mice behaviors. High performance liquid chromatography-fluorimetric detection was performed to determine the concentrations of plasma norepinephrine (NE), dopamine (DA), serotonin (5-hydroxytryptamine, 5-HT). Data were analyzed by two-way analysis of variance using SPSS 16.0. After 53 days of noise exposure, center time and the frequency of line crossing of the exposed mice decreased significantly in the OFT compared with the control group. Meanwhile, transitions and the time spent in the lit compartment of the exposed group decreased significantly in the light/dark box test. After 40 days of HSR noise exposure, the concentrations of plasma DA of the exposed group were significantly higher than those of the control group, while the plasma NE and 5-HT concentrations showed no significant difference between the two groups. The behavioral tests indicate that 70 dB (A) HSR noise can result in anxiety-like behaviors in mice. The physiological results show that plasma DA is more sensitive to HSR noise compared with NE and 5-HT.

  Abstract Polyamines, putrescine, spermidine and spermine, are ubiquitous in living cells and are essential for eukaryotic cell growth. These polycations interact with negatively charged molecules such as DNA, RNA, acidic proteins and phospholipids and modulate various cellular functions including macromolecular synthesis. Dysregulation of the polyamine pathway leads to pathological conditions including cancer, inflammation, stroke, renal failure and diabetes. Increase in polyamines and polyamine synthesis enzymes is often associated with tumor growth, and urinary and plasma contents of polyamines and their metabolites have been investigated as diagnostic markers for cancers. Of these, diacetylated derivatives of spermidine and spermine are elevated in the urine of cancer patients and present potential markers for early detection. Enhanced catabolism of cellular polyamines by polyamine oxidases (PAO), spermine oxidase (SMO) or acetylpolyamine oxidase (AcPAO), increases cellular oxidative stress and generates hydrogen peroxide and a reactive toxic metabolite, acrolein, which covalently incorporates into lysine residues of cellular proteins. Levels of protein-conjuagated acrolein (PC-Acro) and polyamine oxidizing enzymes were increased in the locus of brain infarction and in plasma in a mouse model of stroke and also in the plasma of stroke patients. When the combined measurements of PC-Acro, interleukin 6 (IL-6), and C-reactive protein (CRP) were evaluated, even silent brain infarction (SBI) was detected with high sensitivity and specificity. Considering that there are no reliable biochemical markers for early stage of stroke, PC-Acro and PAOs present promising markers. Thus the polyamine metabolites in plasma or urine provide useful tools in early diagnosis of cancer and stroke.

  Abstract This paper presents a geometric interpolation method for curve approximation in R3 space. Given a curve, the new method is to find an approximation Be}zier curve of degree 4 tangent with the given curve at the two end points and at an inner point as well. The resulting Be}zier curve is explicitly expressed in the parameters of the tangent inner point of both the given curve and the approximation curve. We prove that the approximation order of the new method is 6, which is the optimal approximation order in the traditional conjecture. © 2010 CAD Solutions, LLC.

  Abstract Complex mesh models usually affect the performance of algorithms for CAD applications. Therefore, it is often desirable to use simplified models and at the same time preserves the general features of the shape. In this paper, we present an entropy based surface mesh simplification algorithm which can rapidly and efficiently produce high quality approximations of triangle mesh models. The algorithm uses iterative contraction of edges to simplify models. It first exploits the geometric feature of model surface by estimating vertex curvature entropy and then iteratively contracts edges with relatively lower simplification cost. The simplification cost is measured by a combination of vertex curvature and curvature entropy of the two vertices compositing the edge. The goal is to simplify regions that are relatively flat while preserving salient features of the shape. Experiments show promising results of the algorithm. © 2010 CAD Solutions, LLC.
